New Genetic Discovery Provides Hope in Protecting Against Alzheimer’s Disease

In a groundbreaking development, researchers have made a new genetic discovery that could hold the key to protecting against Alzheimer’s disease. With an estimated 55 million people worldwide suffering from Alzheimer’s and other forms of dementia, finding potential treatments and preventive measures is of utmost importance. This recent discovery offers hope for the future.

The study, led by Joseph Arboleda-Velasquez from Harvard Medical School, focused on a patient with a rare gene mutation called PSEN1-E280A. Individuals with this mutation typically develop mild cognitive impairment at an early age. However, the researchers identified a man who carried this mutation but remained cognitively healthy until his late 60s. This anomaly prompted further investigation.

Neuroimaging studies revealed that the patient had elevated levels of amyloid beta plaques in his cortex, a characteristic of Alzheimer’s disease. However, certain regions of his brain, including the entorhinal cortex responsible for memory, showed lower levels of tau pathology compared to others in the same cohort. This led the researchers to delve deeper into the genetics of this unique case.

The team discovered an unusual variant in the RELN gene, which they named RELN-COLBOS, in the patient’s genome. RELN encodes the protein Reelin, which plays vital roles in brain structure and function. Reelin is involved in neuronal migration and synaptic plasticity, among other processes. This new variant provides valuable insights into signaling pathways that may protect individuals from Alzheimer’s disease.

To understand how the RELN-COLBOS mutation offers protection, the researchers conducted studies using mice. They found that male mice with this mutation exhibited reduced phosphorylation of the protein Dab1, which plays a role in preventing tau hyperphosphorylation. However, this effect was not observed in female mice, suggesting a sex-specific impact. This observation was supported by the patient’s sister, who carried both the PSEN1-E280A and RELN-COLBOS mutations but experienced only partial protection.

While the exact mechanisms behind this protective mutation are not yet fully understood, the findings open up several avenues for further research. Reelin is known to be involved in various neurological and psychiatric disorders, making it a compelling target for potential treatments.

Steven Estus, a molecular geneticist at the University of Kentucky College of Medicine, believes that Reelin is worth exploring further. However, given its vital role in brain development, it is crucial to determine whether the protective effect is a result of early development or a late-stage occurrence. This distinction will guide future efforts in harnessing the potential of this genetic discovery for treatment purposes.

Arboleda-Velasquez emphasizes that the significance of this discovery lies in its promise for patients. The patient’s experience and the findings generated from it provide real-world evidence of the potential of this genetic variant. While more research is needed to fully understand its implications, it offers hope for developing targeted treatments based on individual genetic profiles.
